Abstract

This mini-review seeks to examine the molecular nature of renin-angiotensin system inhibitors and also the way in which their pharmacology relates to clinical effects. This review does not seek to provide an in depth analysis of the extensive studies published which critically review the effects of these inhibitors on blood pressure. Instead these will only briefly be summarised where they highlight advantages or limitations of the drugs discussed within the min-review. Highlighting the various limitations of ACE-inhibitors and angiotensin-II blockers will allow the review to focus on aliskiren, its development, molecular nature and pharmacology. The limitations of aliskiren itself will ultimately be discussed; has aliskiren been successful in cutting off the head of the renin-angiotensin system?
